Breast cancer drug hope for women with Angelina gene

Using the drug olaparib after initial treatment and chemotherapy slashed the risk of cancer returning or spreading by 42 percent. The discovery could lead to more women being cured of high-risk breast cancer after early diagnosis. Dr Simon Vincent, director of research, support and influencing at charity Breast Cancer Now, said: “It’s extremely exciting.

“Olaparib must now be promptly submitted for licensing, and then assessed for use on the NHS, so that women with this type of breast cancer start to benefit.”

The BRCA genes play a role in repairing DNA damage.

In people who carry mutations, mistakes can build up in the DNA, increasing the risk of a cell becoming cancerous. Actress Angelina underwent a double mastectomy in 2013 after discovering she carried a “faulty” BRCA1 gene.

She later had her ovaries and fallopian tubes removed too.

Olaparib is already used to treat some types of ovarian, fallopian tube or primary peritoneal cancer.

It is a type of drug known as a PARP inhibitor and works by disrupting another DNA repair pathway, causing cancerous cells to die.

The international OlympiA trial, involving almost 2,000 patients, is the first to report on olaparib’s ability to stop breast cancer returning when used at an early stage.

After three years, 86 percent of patients treated with the drug were alive and free of invasive breast cancer and second cancers, compared to 77 percent of those who received the usual care and a placebo dose.

Study co-leader Prof Andrew Tutt, of the Institute of Cancer Research and King’s College London, said: “Women with early-stage breast cancer who have inherited BRCA1 or BRCA2 mutations are typically diagnosed at a younger age.

“Up to now, there has been no treatment that specifically targets the unique biology of these cancers to reduce the rate of recurrence, beyond initial treatment such as surgery, hormone treatment, radiotherapy and chemotherapy.”

He estimated that around 1,000 women could benefit from treatment each year in the UK and hundreds of thousands worldwide.

Prof Paul Workman, chief executive of the institute, said: “This is a major breakthrough.”

The findings were presented at the American Society of Clinical Oncology Annual Meeting and published in The New England Journal of Medicine.

Caroline Wheeldon discovered she carries a BRCA2 mutation after finishing treatment for early breast cancer.

She was diagnosed with stage one triple negative breast cancer in 2019 after pains alerted her to a lump.

Caroline, 40, said: “When I found out I had cancer, my world fell apart.

“I knew straight away that I wanted to have a double mastectomy to reduce my risk of it returning.”

She had a unilateral mastectomy followed by chemotherapy. Due to her age and cancer type, she also underwent genetic testing which revealed she had the BRCA2 mutation, greatly increasing her risk of certain cancers.

She was the first person in her family to have had cancer. But her 70-year-old mother has been discovered to be a carrier of the mutation.

She had a second mastectomy in August last year and had her ovaries and fallopian tubes removed in December 2020. Caroline lives in Barnsley with her husband and two children, Aleisha, 11, and Leo, nine.

She said today’s breakthrough gave her hope that better treatments will be available in the future.

She said: “It’s so reassuring to know about the possibility of this new treatment.

“I’m really pleased that it might be available by the time my children are grown up, and to help anyone who’s diagnosed in the future.”


A drug that acts “like a guided missile” to target prostate cancer can extend the lives of men with advanced disease, a study suggests.

The treatment detects the presence of a molecule called prostate-specific membrane antigen (PSMA) on the surface of tumour cells, before delivering a dose of radiation to kill them.

The “seek and destroy” method can shrink tumours while leaving healthy cells unharmed.

An international trial involved 831 patients with advanced prostate cancer, meaning it had spread to other parts of the body.

Those treated with the drug typically survived for 15.3 months compared with 11.3 for those who received standard care alone.

It also extended the average time that passed before patients’ cancers got worse from 3.4 months to 8.7 months.

It is estimated that between a third and half of the 10,000 men diagnosed with advanced prostate cancer each year have tumours with high levels of PSMA and could benefit.

Study co-author Professor Johann de Bono, of The Institute of Cancer Research in London, said: “I believe that these results can change the standard of care for some men with advanced prostate cancer.”

Dr Matthew Hobbs, at Prostate Cancer UK, said the results were “hugely exciting”.

Eternals trailer: Angelina Jolie and Richard Madden star in millennia-spanning epic WATCH

For the first time, this year will see four Marvel Studios movies released in cinemas, beginning with Black Widow and Shang-Chi and the Legend of the Ten Rings. But now the first teaser trailer for the third film Eternals has dropped promising a millennia-spanning epic. The MCU blockbuster follows an immortal alien race created by the Celestials who have secretly lived on Earth for over 7000 years without interfering in events.
However, following a tragedy in the aftermath of Avengers Endgame, the Eternals are reuniting to protect humans from their evil counterparts known as the Deviants.

Angelina Jolie stars as Thena, a fierce warrior who can form weapons out of cosmic energy.

While Richard Madden plays Ikaris, the tactical leader of the superhero team who has incredible strength, is able to fly and can shoot beams from his eyes.

From the trailer, it’s clear he has a special bond with Gemma Chan’s Sersi, who Marvel boss Kevin Feige has described as the film’s lead.

Spider-Man: No Way Home will star Tom Holland’s Web-Slinger in his third solo outing, with Benedict Cumberbatch’s Doctor Strange mentoring him.

The upcoming movie is heavily rumoured to feature former Sony Peter Parkers Tobey Maguire and Andrew Garfield, plus a whole host of Spider-Man villains from across the multiverse.

It’s not clear yet how the Eternals will crossover with the Avengers, but with alternate realities set to open up no doubt their help will be needed at some point.

Perhaps they’ll next show up in 2022’s Doctor Strange in the Multiverse of Madness or even Thor: Love and Thunder?

Also set for release in 2022 are Black Panther: Wakanda Forever and Captain Marvel sequel The Marvels.

While 2023 will see Ant-Man and the Wasp: Quantumania and Guardians of the Galaxy Vol 3 land in cinemas.

Also in the works are Blade, Fantastic Four, Deadpool 3 and the mysterious Avengers 5.

Eternals is released in cinemas on November 5, 2021.

As for when Angelina Jolie will be “directing a movie again,” we’re hearing that the actress has a loose timeline in mind. The 45-year-old Maleficent star recently made it known that directing is where her true passions lie — something that became impractical after a “change in [her] family situation,” which was a hint at her split with Brad Pitt in 2016 — while speaking to Entertainment Weekly. While the mother of six admitted that this is why she “went back to doing a few acting jobs” so she could “be home more,” we learned what Angelina’s upcoming career plans are now that she is working out these “issues” with Brad.

“Since the moment she became a mom Angelina has always put her kids first. They come before anything and anyone. And with the divorce they have needed so much more of her time and attention so, as she said, taking on a big directing project just has not been feasible,” a source close to Angelina EXCLUSIVELY tells HollywoodLife, echoing what the actress herself said in her new interview. Angelina has not directed since the release of her 2017 feature, First They Killed My Father. However, “it is absolutely something she will tackle again,” our source clarifies, who adds, “She loves it but only when she can really give her all and right now her focus is still first and foremost on being a mom.”
So, what about right now? “She still wants to work but, as far as taking on an all-consuming project like directing a movie, she will wait until she feels the kids are ready for that. Until they are older,” our source further reveals, but adds, “But it won’t be that long, they’re all teenagers now.” Angelina and Brad’s youngest children — twins Vivienne and Knox — are now 12 years old, while Shiloh is 14, Zahara is 16, Pax is 17 and Maddox is already 19 and a college student.

“Angelina knows these years go by in the blink of an eye so it’s not just about what’s best for her kids,” our source continues. “She also doesn’t want to miss her time with them. She is savoring her time with her kids as much as she can, so she has no regrets about putting off directing a big project.” Indeed, Angelina takes bonding time with her children seriously — she’s often spotted out and about on shopping trips in Los Angeles with them!

“Her kids are the ultimate focus of everything, and she always emphasizes how grateful she is for every moment they agree to spend with her,” our source adds. “Like I said, they are teenagers now, so they are already starting to go all their own ways. They have their friends and all their activities, plus their schoolwork. They’re all very busy, so she doesn’t take it for granted that they still love spending time with her.”

While Angelina confessed that the end of her marriage did affect the jobs she was taking, “for the most part, her career and life are completely separate from her issues with Brad,” a second source, who’s also close to the Girl, Interrupted star, EXCLUSIVELY tells HollywoodLife. However, our second insider does say that these issues (i.e. their ongoing divorce case and custody battle) “have played a small part in her career trajectory in the last year.”

“It has just made things have an extra step to get completed. It takes a lot to be a super mom,” this source adds, referring to the loose ends Angelina and Brad have had to tend to over the past year. However, our source reveals that Angelina’s looking forward to the next chapter in her life by adding, “She is really looking forward to getting back on track though by the end of the year with her personal and professional life and return to the Angie she always has been. Her issues with Brad, though complicated, is not going to consume her in the near future. It is just something she must achieve for her own peace of mind and to move forward rather than backwards.”

The close bond between Angelina Jolie and her daughter, Vivienne Jolie-Pitt, was apparent in this sweet photo of their shopping outing in Hollywood on March 29.

Angelina Jolie[1] and Vivienne Jolie-Pitt[2] have two things in common, besides their last name: a love for shopping and one another! The 45-year-old Maleficent star and her 12-year-old daughter were seen on another shopping excursion, this time in Hollywood, on March 29. As they left a Forever 21 store, they were seen with their arms sweetly wrapped around one another in a half-embrace.

Angelina opted for a business-casual ensemble for the mother-daughter outing: a cinch-tie trench coat and black slide-on sandals[3]. Meanwhile, her mini-me opted for a trendy oversized sweater with a dainty floral graphic and athletic sneakers. Vivienne was nearly as tall as her famous mother in the photo!

Angelina and Vivienne really do love to shop together. Just three days before this outing, they were seen leaving a fabric store[4]. Angelina wore a similar longline coat that time (beige, minus the tie) while Vivienne also rocked a sporty streetwear look for the outing.

As you can see, Angelina is hanging out with her kids per usual amid a custody battle playing out with her estranged husband Brad Pitt[5]. Angelina accused her 57-year-old ex, whom she filed for divorce from in Aug. 2016, of domestic violence in court documents[6] filed on March 12. She also claimed to have “proof and authority” to back up this allegation; AKA, potential “testimony” from her children (Vivienne, in addition to her five other minor children Pax, 17, Zahara, 16, Shiloh, 14, and Knox, 12). Angelina and Brad’s eldest child, Max, is 19 years old and therefore a legal adult — he is currently studying in South Korea.
This development in Angelina and Brad’s years-long custody drama[7] happened a few months after Brad requested full 50/50 custody of the children in Oct. 2020. This was a shakeup to their prior temporary agreement, which was reported in Nov. 2018. Brad had reportedly been awarded less than 50 percent of custody, and would “see an increase in his custodial time with the kids,” The Blast[8] reported at the time.

Vivienne, 12, Shows She’s Nearly As Tall As Mom Angelina Jolie While Out On Shopping Trip

Vivienne Jolie-Pitt visited a fabric store with Angelina Jolie and rocked a ponytail just like her older sister Shiloh.

Angelina Jolie[1], 45, and her youngest daughter Vivienne Jolie-Pitt[2], 12, were spotted getting in a shopping trip[3] on Mar. 26 and it proved just how much the latter is growing! The actress; mini-me looked almost as tall as her mom while wearing a black hoodie, white socks, and black and white Nike sneakers. She also had her hair up in a bun similar to the one her sister Shiloh, 14, sometimes wears.

Angelina looked gorgeous in a long white dress under a long tan trench coat and white slip-on shoes. She had her hair down and appeared relaxed as she followed her daughter, who was pushing a cart outside. They both also wore face masks.

Angeline and Vivienne’s outing comes a week after the Girl Interrupted star made headlines for accusing her ex-husband and father of her six kids, Brad Pitt[4], 57, of domestic violence. In leaked documents[5] released earlier this month, Angelina claims to have “proof and authority” to support her allegations and a source told us about the situation.

“This deliberate leak of this information is to no one’s benefit and is simply the continuation of an ongoing relentless effort to target and tarnish Brad,” the source EXCLUSIVELY told HollywoodLife. “Over the past four and a half years since the divorce filing there have been a never ending series of allegations, which seem to be solely to elevate one side by taking down the other. Hopefully some sanity will prevail in the future.” [6]

Another leaked document in the case shows that Angelina seems to be bringing her and Brad’s children into the court battle[7]. It says she has “proof and authority in support thereof RE: testimony of minor children” on her behalf and refers to Pax, 17, Zahara, 16, Shiloh, 14, and Vivienne and her twin brother Knox, 12. Their oldest son Maddox is already a legal adult at the age of 19.

Angelina and Brad were married in 2014 and split just two years later. Since then, they have been involved in a custody battle regarding their children and these latest leaked documents prove that they’re still privately going through disagreements.
